Delineating the key virulence factors and intraspecies divergence of Vibrio harveyi via whole-genome sequencing

چکیده

Vibrio harveyi is one of the major pathogens in aquaculture. To identify key virulence factors affecting pathogenesis V. towards fish, we conducted a field investigation for three representative fish farms infected with harveyi. Multilocus sequence typing (MLST) and whole-genome sequencing were to delineate phylogenetic relationship genetic divergence A total 25 strains isolated from diseased groundwater subtyped into 12 types by MLST. Five genes, mshB, pilA, hutR, ureB, ureG, variably presented sequenced strains. The gene profiles strongly correlated distinct pathogenicity strains, strain harboring all five genes exhibiting highest fish. Phenotype assay confirmed that reduced decreased motility biofilm formation ability. Additionally, type VI secretion system, namely T6SS1, T6SS2, T6SS3, identified which can be classified six, four, subtypes, respectively. In conclusion, results indicated level mainly determined above may play vital roles environmental adaptation

Complete genome sequence of virulence-enhancing Siphophage VHS1 from Vibrio harveyi.

Vibrio harveyi siphophage 1 (VHS1) is a tailed phage with an icosahedral head of approximately 66 nm in diameter and an unornamented, flexible tail of approximately 153 nm in length. When Vibrio harveyi 1114GL is lysogenized with VHS1, its virulence for the black tiger shrimp (Penaeus monodon) increases by more than 100 times, and this coincides with production of a toxin(s) associated with shr...

Delineating the Origins of Vibrio parahaemolyticus Isolated from Outbreaks of Acute Hepatopancreatic Necrosis Disease in Asia by the Use of Whole Genome Sequencing

Acute hepatopancreatic necrosis disease (AHPND) is an emerging penaeid shrimp disease caused by Vibrio parahaemolyticus. Although V. parahaemolyticus has been isolated and sequenced from several Asia countries, the epidemiological links among the AHPND outbreaks in different locations remain unclear. In this study, we sequenced the genomes of nine strains isolated in China between 2014 and 2016...

Comparing whole genome sequencing and exome sequencing

Introduction Next-generation DNA sequencing empowers scientists to identify genetic variations associated with human disease at higher resolution and greater sensitivity than previously possible. Two approaches are commonly employed -exome sequencing and whole genome sequencing. Exome sequencing targets protein-coding regions comprising approximately 1% of the human genome, while whole genome s...

Whole exome and whole genome sequencing.

PURPOSE OF REVIEW The purpose of this review is to describe the new DNA sequencing technologies referred to as next-generation sequencing (NGS). These new methods are becoming central to research in human disease and are starting to be used in routine clinical care. RECENT FINDINGS Advances in instrumentation have dramatically reduced the cost of DNA sequencing. An individual's entire genome ...

Contrasting Inter- and Intraspecies Recombination Patterns in the “Harveyi Clade” Vibrio Collected over Large Spatial and Temporal Scales

Recombination plays an important role in the divergence of bacteria, but the frequency of interspecies and intraspecies recombination events remains poorly understood. We investigated recombination events that occurred within core genomes of 35 Vibrio strains (family Vibrionaceae, Gammaproteobacteria), from six closely related species in the so-called "Harveyi clade." The strains were selected ...
